The Primary Information of Voltage-gated Proton Channel
1. Summary
The voltage-gated proton channel Hv1 is a member of the family of voltage-gated ion channels [1]. Hv1 channel plays roles in the regulation of reactive oxygen species (ROS) production by the NADPH oxidase and cellular pH homeostasis [2].
The VSDs of voltage-gated ion channels are made of four transmembrane segments (S1 through S4). A highly conserved phenylalanine is located in the S2 segment of the VSD and acts as a part of the "gating charge transfer center" [3].
2. Binding Sites
Blocker
The highly conserved phenylalanine of the human Hv1 channel (hHv1) is F150 (red sphere in the 3D Structure Viewer), which affects the strength of the interaction between the Hv1 channel and the blocker 2GBI [4].
The voltage-gated proton channel (Hv1/VSOP) is inhibited by Zn2+, of which the binding site is located in the extracellular region (residue His189, Asp119, His136 blue sphere in the 3D Structure Viewer) [5].
